Job Description
PineTree Therapeutics is actively seeking an experienced and motivated Principal Scientist to join our discovery team. This position will play a critical role in antibody discovery utilizing 3D structural modelling information of antigen or antibody-antigen complex as a guide. The ideal candidate will actively support monoclonal and multi specific therapeutic antibody selection for further development. The position should have broad experience of the real-world complications of therapeutic biologics development as a way to de-risk programs. Extensive experience/knowledge of analytical method development, customized multi specific antibody purification, developability assessment and lead selection would be a plus. This is an onsite role in the greater Cambridge MA area.
Duties and Responsibilities
- Structural or 3D modelling (or AI-aid) based rational design to support de novo therapeutic antibody discovery and lead optimization.
- Multi specific antibody optimization and in-silico developability risk assessment to support lead modality selection
- In silico prediction to guide the purification and analytical strategy utilizing modern techniques, including but not limited to CEX-FPLC, AEX-FPLC affinity chromatography, RP-HPLC, HIC-HPLC, CEX-HPLC and LC/MS et al
- Support asymmetric multi specific antibody purification and analytical analysis for lead selection at different stages
- Lead projects from discovery to late-stage development including design of experiments, scheduling, setting up, executing, troubleshooting, and writing reports
- Write, review and revise research batch records, protocols, reports and presentations
- Train and mentor junior scientist(s)
Qualifications
- PhD or advanced degree in Structure biology, Computational structural biology, Chemical engineering, Bioengineering, Biotechnology, Chemistry, Biochemistry or other related fields
- 5+ years of experience in postdoctoral, biotech or pharmaceutical industries specifically focused on Protein/Antibody structural biology, Protein/Antibody 3D modelling (or AI-aid), Computational modelling, Machine learning, Multi specific antibodies purification or analytics et al
- Proficiency of modern computational structure tools such as Alpha-Fold, MOE, Schrodinger or RoseTTAFold et al
- Experience/knowledge in affinity chromatography using ion-exchange, mixed-mode, and hydrophobic resins et al
- Excellent organizational skills with the ability to work on multiple projects at a time.
- Strong technical writing skills
- Excellent communication skills and presentation skills
- The title will be adjustable based on candidate’s experience
- Prior experience in Biotech, pharmaceutical industry, PhD/postdoc training in structural biology, chemical engineering or protein/antibody engineering
